Cities: Skylines to klasyczny przedstawiciel strategii typu city builder. Celem rozgrywki jest wybudowanie sprawnie funkcjonującego miasta przy jednoczesnym zachowaniu jego płynności finansowej. Całość należy zaprojektować od podstaw – gracz zaczyna na obszarze o rozmiarze 4 km kwadratowych (z czasem może go powiększyć do maksymalnie 36 km kwadratowych) i musi poprowadzić na nim drogi, rozmieścić strefy mieszkalne, handlowe i przemysłowe, zaprojektować trasy komunikacji miejskiej oraz dbać o zadowolenie ludzi. Kolejne elementy związane z zarządzaniem miasta odblokowywane są stopniowo, wraz ze wzrostem populacji.

Do dyspozycji gracza przygotowano kilka typów dróg, które zapewniają różną przepustowość. Wszystkie (poza autostradami) umożliwiają również stawianie przy nich różnych budynków oraz stref mieszkalnych, handlowych i przemysłowych. Jeśli dana ulica będzie za wąska, by zagwarantować płynny przepływ pojazdów, pojawią się korki utrudniające transport towarów i przemieszczanie się ludzi.
